This recombinant whole-cell biocatalyst expresses a four-enzyme cascade — 7α-HSDH, 7β-HSDH, LDH, and GDH — for the efficient biotransformation of chenodeoxycholic acid (CDCA) into ursodeoxycholic acid (UDCA) or tauroursodeoxycholic acid (TUDCA). Developed using E. coli, the system enables high-yield enzymatic conversion (>98%) with in vivo cofactor regeneration, eliminating the need for external NAD(P)H. The cascade includes: 7α-HSDH: Oxidizes CDCA to 7-keto-LCA 7β-HSDH: Reduces 7-keto-LCA to UDCA LDH & GDH: Regenerate NAD⁺ and NADPH for continuous enzyme cycling This scalable platform is ideal for pharmaceutical manufacturers, CDMOs, and research teams working on bile acid derivatives, steroid APIs, or sustainable biocatalysis. Available as a live recombinant strain or lysate.
